Its simply the 2 versions the manifold 'physically'come in;
4-2-1; where the manifold bolts onto the head there are 4 pipes which when you follow the manifold down just towards the point in which it bends under the sump you will notice it merges into 2 pipes (bottom end area) and then if you follow it to where it goes to bolt onto the centre pipe it merges into 1 pipe.
And the 4-1 is basically the same manifold but instead of merging into 2 pipes, just merges into 1 from 4 near the bottom of the manifold
So think of it as a physical description.
4-1 manifold
iirc provide more top end 'flow' than the 4-2-1 but not as much low end torque gains as the 4-2-1
